Heart benefits of two diabetes drug types: GLP-1 receptor activators versus SGLT2 inhibitors

Abstract

The study included 1,245,211 adults with type 2 diabetes mellitus receiving metformin who initiated second-line therapy with glucagon-like peptide 1 receptor agonists or sodium-glucose cotransporter 2 inhibitors.

  • Individual GLP-1 receptor agonists and SGLT2 inhibitors showed broadly similar cardiovascular effectiveness.
  • Semaglutide and empagliflozin had comparable risks for major adverse cardiovascular events (3-point MACE) and expanded outcomes (4-point MACE).
  • Meta-analytic hazard ratios for semaglutide and empagliflozin were 1.05 for 3-point MACE and 0.95 for 4-point MACE.
  • Findings were consistent within the subgroup of patients with established cardiovascular diseases.
  • Study diagnostics confirmed sufficient statistical power to detect similarity in hazard ratios between 0.8 and 1.2.
