Abstract

Bacteroidetes abundance was significantly reduced in unmedicated adolescents with depression.

  • Key indole derivatives, including IAA, IPA, and IAld, were markedly decreased in depressed adolescents and negatively correlated with depression severity scores.
  • In adolescent mice with depression induced by chronic restraint stress, levels of IAA, IPA, and IAld also declined.
  • A high-tryptophan diet and direct supplementation with indole derivatives substantially improved depression-like behaviors in these mice.
  • Conversely, a low-tryptophan diet induced depressive behaviors in otherwise healthy mice.
  • Microglial cells in mice with chronic restraint stress showed features of overactivation, which were restored by high-tryptophan diet and indole derivative supplementation.
  • Indole derivatives activated AhR signaling, balancing pro-inflammatory and anti-inflammatory factors, thus promoting neurogenesis and remodeling microglial morphology.
