A New Role for Liver Cell MLKL in Causing Mitochondrial Problems and Aging in the Liver

Updated

Abstract

MLKL is elevated in aged hepatocytes and may drive liver aging through non-necroptotic mechanisms.

  • Necroptosis, a form of inflammatory cell death, is activated in aging livers but does not occur with MLKL overexpression in hepatocytes.
  • MLKL overexpression in hepatocytes leads to increased levels of markers associated with cellular aging, such as p16INK4a and p21WAF1/Cip1.
  • Mitochondrial dysfunction is induced by MLKL, characterized by impaired respiration and increased reactive oxygen species.
  • The mitochondrial stress caused by MLKL is linked to the release of pro-inflammatory extracellular vesicles and promotes senescence in both hepatocytes and non-parenchymal cells.
  • Macrophages are identified as a population enriched in senescence, suggesting that liver aging may be amplified through signaling from surrounding cells.
