Creating Herbicide-Resistant Alfalfa Using Precise Gene Editing

Abstract

Alfalfa lines were successfully generated that are resistant to nicosulfuron and haloxyfop-P-methyl through multi-gene prime editing.

  • A highly optimized prime editing system was developed specifically for alfalfa.
  • Mutations were introduced in three target genes, MsALS1, MsALS2, and MsACC1, to confer herbicide resistance.
  • Mutations in MsALS1 and MsALS2 are aimed at providing resistance to ALS-inhibiting herbicides.
  • Mutations in MsACC1 target resistance to aryloxyphenoxypropionate (APP) herbicides.
  • The Csy4 nuclease system facilitated simultaneous editing of all three genes.
