High-Precision Virus-Like Particles Carrying Cas12m for Changing DNA Bases from A to G and C to T

Updated

Abstract

Shortening the linker in dual base editors can achieve single-nucleotide substitutions with less than 0.8% bystander activity.

  • High editing precision is possible by engineering the linker between the Cas12m domain and the deaminase domain of the base editor.
  • Different linker sequences were tested to create dual base editors with narrowed editing windows.
  • The optimized dual base editors can edit a single nucleotide at a target position with high accuracy.
  • The high-precision base editor retains up to 90% of the editing efficiency of the original version.
  • These advancements may benefit therapeutic approaches where precision is essential and off-target effects are undesired.
