Abstract

PER1 knockout rats exhibited synaptic potentiation that decayed to baseline levels within approximately 90 minutes during both light and dark phases.

  • In wildtype rats, long-term potentiation (LTP) induced by theta-burst stimulation (TBS) was consistently greater during the dark phase compared to the light phase.
  • Wildtype rats expressed long-term synaptic depression (LTD) following low-frequency stimulation (LFS) during the dark phase but not during the light phase.
  • In contrast, PER1 knockout rats were able to induce LTD during both the dark and light phases following LFS.
  • Both LTP and LTD in wildtype and PER1 knockout rats were dependent on the activation of N-methyl-d-aspartate receptors.
  • Basal synaptic transmission was reduced, and GluA1 S845 phosphorylation was increased during the dark phase in both wildtype and PER1 knockout rats.
