Functional Groups Linked to Olaparib Resistance Beyond DNA Repair Identified by CRISPR Screens

Updated

Abstract

Eight functional modules associated with PARP inhibitor resistance were identified through a comprehensive analysis of knockout screens.

  • Genes related to DNA-damage repair and replication stress were found to be recurrently associated with PARP inhibitor resistance.
  • Cell-cycle regulation and chromatin remodeling were also identified as significant pathways contributing to resistance.
  • Mitochondrial function genes emerged as recurrent resistance hits, although their role in olaparib resistance remains poorly explored.
  • Resistance is suggested to be a complex and adaptive response influenced by various cellular programs.
  • The analysis emphasizes the need for further validation of candidate resistance drivers and informs potential combination therapy strategies.
