Homoplantaginin improves osteoarthritis by activating cell cleanup and reducing inflammation in cartilage cells

Abstract

Homoplantaginin (Homo) significantly alleviated OA-related pathological manifestations in both in vivo and in vitro models.

  • Homo reduced cartilage degradation and normalized extracellular matrix metabolism.
  • Chondrocyte viability was enhanced, along with suppression of apoptosis and oxidative stress.
  • Transcriptomics analysis identified mitophagy as a potential mechanism of action for Homo.
  • Sirt3 was found to be a direct target protein of Homo, which may play a key role in its therapeutic effects.
  • Homo activates the Sirt3/PINK1/Parkin signaling pathway, promoting mitophagy and restoring mitochondrial function.
  • Rescue experiments confirmed the therapeutic efficacy of Homo in ameliorating OA progression.
