hTERT Immortalization Keeps Human Umbilical Cord Stem Cells Stable and Preserves Their Small Vesicles’ Ability to Regulate Immunity and Activate Early Egg Follicles

Abstract

Overexpression of human telomerase reverse transcriptase (hTERT) significantly alleviated senescence-related deterioration in human umbilical cord mesenchymal stromal cells (hUCMSCs).

  • Late-passage hUCMSCs exhibited canonical senescence features, including impaired proliferation and mitochondrial dysfunction.
  • Small extracellular vesicles (sEVs) derived from senescent hUCMSCs showed diminished immunomodulatory activity.
  • hTERT expression largely restored the immunomodulatory function of sEVs from senescent cells.
  • Senescence was linked to altered sEV cargo, particularly an increase in miR-217-5p, which reduced immunomodulatory potency.
  • H11-sEVs from hTERT-expressing cells retained a young-like protein cargo profile associated with immune regulation.
  • In aged female mice, H11-sEVs demonstrated protective effects on ovarian function.
