A human breast cancer model using a new test to study telomerase-blocking drugs

Abstract

Telomerase activity varies markedly across 39 human cancer cell lines, with MCF7 cells exhibiting 33.8% of the activity seen in HeLa cells.

  • A new method for detecting telomerase activity using a non-amplified primer extension assay was developed, improving characterization and quantification.
  • Telomerase activity was found to be significantly different among various human breast cancer cell lines.
  • No correlation was observed between telomerase activity and telomere length, indicating alternative mechanisms may be involved in telomere maintenance.
  • MCF7 and MX1 cell lines were identified as suitable models for testing agents interacting with telomeres and telomerase.
  • Cisplatin and TMPyP4 treatment resulted in down-regulation of telomerase activity in both MCF7 cells and MX1 tumor xenografts.
