Testing human heart cells finds improved fat particles for gene editing in the heart

A human cardiomyocyte screen identified LNPs that can deliver gene-editing cargo to heart models, with strongest cardiomyocyte delivery after direct injection.

Evidence

This preclinical platform study screened LNPs in hiPSC-CMs, tested Cre mRNA delivery by four routes in Ai14 mice, and tested ABE correction in DMD patient-derived hiPSC-CMs plus a humanized DMD mouse model.

Caveat

In vivo cardiomyocyte transfection was route-dependent and strongest at intramyocardial injection sites, so the abstract does not establish systemic human cardiac delivery or clinical benefit.
