Aging cell

A new human cell model for studying normal aging and finding anti-aging treatments

Updated

Abstract

Essence

A human trophoblast stem-cell-to-syncytiotrophoblast system is proposed as an accelerated cellular model for normal aging and anti-aging screening.

Evidence

Cell-model experiments with in vitro hTSC-derived STBs showed CTB-STB maturation and aging-like hallmarks, plus attenuation of senescence signals by mTOR inhibitors and senolytics.

Caveat

The model uses placental cell differentiation and readouts, so it does not by itself demonstrate organism-level anti-aging effects.

What this is

  • This research introduces a human cellular model for studying aging through trophoblast stem cells (hTSCs) differentiating into syncytiotrophoblasts (STBs).
  • The model mimics key aging features observed in vivo, such as and genomic instability.
  • It also facilitates the evaluation of anti-aging interventions, bridging fundamental aging research with clinical applications.

Essence

  • The hTSC-STB system effectively replicates human cellular aging features, providing a novel platform for investigating aging and testing anti-aging therapies.

Key takeaways

  • The hTSC-STB model accurately mirrors in vivo aging processes, demonstrating features like DNA damage, cell cycle arrest, and ().
  • Known anti-aging agents, including mTOR inhibitors and senolytics, effectively reduce senescence markers in STBs, indicating their potential for therapeutic applications.
  • The system allows for scalable screening of anti-aging compounds, identifying novel candidates that could impact aging processes in human tissues.

Caveats

  • The placenta's transient nature may limit the model's applicability to long-lived tissues, potentially affecting the generalizability of findings.
  • The reliance on hCG and CGA levels as markers for aging may not fully encompass all aging hallmarks, necessitating additional validation.
  • Candidates identified in this system require further validation in diverse models to ensure their relevance in broader aging contexts.

Definitions

  • cellular senescence: A state where cells stop dividing but remain metabolically active, contributing to aging and tissue dysfunction.
  • senescence-associated secretory phenotypes (SASPs): A collection of pro-inflammatory cytokines secreted by senescent cells that can influence neighboring cells and promote aging.
