Simple One-Step Test to Detect Human Influenza B Virus Using DNA Amplification and CRISPR Technology

Abstract

The one-step detection method can identify human influenza B virus at concentrations as low as 2.8 copies/μL within 25 minutes.

  • The optimal reaction conditions for the method include a temperature of 37 ℃ and specific concentrations for Cas12a, ssDNA probe, and primers.
  • High specificity was achieved, with no cross-reaction noted for other respiratory viruses, including SARS-CoV-2 and influenza A (H1N1).
  • Clinical testing demonstrated a sensitivity of 93.33% and a specificity of 100% compared to RT-qPCR results.
  • The entire detection process, from nucleic acid release to fluorescence signal output, can be completed in 30 minutes.
