Making human regulatory T cells resistant to the drug tacrolimus by editing the FKBP12 gene

Abstract

Tacrolimus significantly reduces the proliferation of control regulatory T cells, even in high IL-2 conditions.

  • FKBP12 deletion in human regulatory T cells prevents the antiproliferative effects of tacrolimus.
  • Control Tregs showed reduced proliferation when treated with 10 ng/mL tacrolimus, regardless of IL-2 concentration.
  • FKBP12-knockout Tregs maintained proliferation levels similar to untreated cells.
  • No significant changes in Treg phenotype or stability were observed after FKBP12 deletion or tacrolimus treatment.
  • Edited Tregs retained normal expression of the FOXP3 marker and a transcriptomic profile similar to control Tregs.
