Abstract

Essence

PEG-free hydrophilic polymer mRNA-LNPs, especially POEGMA(+) LNPs, showed less repeat-dose accelerated blood clearance in a mouse model.

Evidence

This preclinical formulation study compared POEGMA, PDMA, and PHPMA polymer LNPs in a mouse repeated-dose clearance model after two weekly doses and in the presence of anti-PEG antibodies.

Caveat

The work measures short-term LNP clearance in mice, not therapeutic efficacy, long-term repeat dosing, or human anti-PEG antibody responses.
