Low oxygen changes daily rhythm gene activity in liver cancer

Abstract

Treatment with CoCl2 at 50, 100, or 200 µM resulted in concentration-dependent upregulation of hypoxia-inducible factors in liver cancer cells.

  • The protein levels of HIF-1α and HIF-2α increased in a dose-dependent manner following CoCl2 treatment.
  • Circadian rhythm gene expression was altered, with increased levels of Clock, Bmal1, and Cry2, while Per1, Per2, Per3, Cry1, and CKIε levels decreased.
  • Transfection with HIF-1α plasmid also resulted in increased Clock, Bmal1, and Cry2 levels, with decreased expression of other circadian genes.
  • Transfection with HIF-2α plasmid led to upregulation of Clock, Bmal1, Per1, Cry1, Cry2, and CKIε, while downregulating Per2 and Per3.
  • A hypoxic environment may disturb circadian gene expression in hepatocellular carcinoma, with HIF-1α and HIF-2α playing roles in this disturbance.
