A significant decrease in CCN2 expression was observed in hypoxic nucleus pulposus cells.

  • Putative hypoxia response elements are identified in both human and mouse CCN2 promoters.
  • HIF-1α appears to regulate CCN2 expression in a way that is not directly linked to the identified hypoxia response elements.
  • Silencing CCN2 leads to increased levels of HIF-1α and its target gene expression.
  • Treatment with recombinant CCN2 reduces the ability of HIF-1α to activate its target genes.
