Abstract

HuR regulates key extracellular matrix genes in nucleus pulposus cells.

  • HuR silencing did not change levels of Hif-1α or its target genes in nucleus pulposus cells.
  • RNA-Sequencing revealed that HuR affects the expression of several important extracellular matrix genes.
  • Decreased secreted collagen I and Sdc4, along with increased pro-MMP13, were observed in HuR-knockdown cells.
  • HuR was found to bind to Tgf-β3 and Sdc4 mRNAs, indicating a direct regulatory role.
  • Significant decreases in extracellular acidification rate and mitochondrial oxygen consumption were noted in HuR-silenced nucleus pulposus cells.
