Abstract

HIF-1α is essential for the expression of carbonic anhydrase isoforms CA9 and CA12 in intervertebral disc cells.

  • Intervertebral disc degeneration is linked to chronic low-back pain and relies on the health of the nucleus pulposus.
  • Analysis showed that carbon dioxide hydration is a significant source of extracellular proton production in nucleus pulposus cells.
  • Nucleus pulposus cells expressed CA9 and CA12 isoforms, which are induced under low-oxygen conditions.
  • HIF-1α was found to bind to specific regions on the CA9 and CA12 gene promoters, enhancing their expression.
  • Inhibition of carbonic anhydrase activity reduced the ability of cells to maintain intracellular pH and decreased cell viability.
  • Bicarbonate buffering through the HIF-1α-carbonic anhydrase axis may be crucial for cell survival in the intervertebral disc's hypoxic environment.
