Immune memory influences antibody responses to KP.2 and LP.8.1 vaccine doses

Abstract

A single KP.2 vaccine dose generated durable, cross-reactive antibody responses for up to six months in 21 individuals.

  • KP.2 vaccination increased spike-specific binding and neutralizing antibodies against the ancestral WA1 strain and other variants.
  • Half-lives for WA1- and KP.2-specific IgG responses were estimated at 770 and 248 days, respectively.
  • The vaccine induced a greater increase in IgG1 and IgG4 subclasses than in IgG2 and IgG3 responses.
  • Most antibodies elicited by the vaccine were cross-reactive, maintaining broad neutralizing activity against several Omicron subvariants.
  • Neutralization assays showed a 4.2-fold reduction in neutralizing titers against variant XFG.3.12 relative to KP.2.
  • An additional LP.8.1 vaccine dose modestly increased neutralizing activity against some divergent variants while preserving cross-reactivity.
