High drug-loading lipid nanoparticles (hDDLN) were developed to improve cisplatin delivery and efficacy.
- Multicarboxyl dendritic lipopeptides were created to enhance cisplatin loading through platinum coordination.
- DG3 was identified as the optimal lipopeptide, resulting in LNPs with good stability and delivery efficiency.
- Pt/hDDLN demonstrated a superior therapeutic index compared to conventional liposomes in both laboratory and animal studies.
- Combining Pt/hDDLN with STING agonists improved immunotherapy by increasing cytotoxic T lymphocyte infiltration and promoting dendritic cell maturation.
