Lipid nanoparticles were developed to enhance mRNA delivery and elicit potent tumor-specific T cell responses.

  • Inefficient mRNA delivery and inadequate dendritic cell activation limit the efficacy of mRNA-based cancer vaccines.
  • Cold tumors exhibit reduced effectiveness due to poor innate immune activation and antigen presentation.
  • Codelivery of STING agonists with tumor antigen-encoding mRNA in lipid nanoparticles enhances immune activation.
  • This strategy activates type I interferon signaling and upregulates costimulatory molecules.
  • Results indicate superior antitumor efficacy through improved antigen presentation and T cell responses.
