Hyperactivation of the cAMP-PRKA/PKA signaling axis is associated with impaired mineralization in fibrous dysplasia.

  • Impaired osteogenesis of bone marrow stromal cells is central to the pathogenesis of fibrous dysplasia.
  • Disruption of mitochondrial homeostasis occurs through hyperactivation of cAMP-PRKA/PKA signaling.
  • Enhanced mitochondrial fusion leads to an elevated mitochondrial membrane potential and impaired mitophagy.
  • Diminished secretion of amorphous calcium phosphate is observed, resulting in mineralization failure.
  • The mechanism involves PRKA activation inhibiting the recruitment of DNM1L/DRP1 to mitochondria, suppressing mitochondrial fission.
