Computer Modeling of Gene Editing to Restore Factor VII Production in Inherited Deficiency

Updated

Abstract

Computational modeling suggests that targeting HNF4α-associated regulatory regions could increase simulated F7 transcription toward approximately 87% of wild-type levels.

  • Hereditary Factor VII deficiency is linked to reduced F7 gene expression and impaired blood coagulation.
  • Six high-confidence HNF4α binding motifs were identified within the proximal F7 promoter, some overlapping known disease-associated variants.
  • Liver expression analyses indicated positive correlations between F7 and the transcription factors HNF4A (r = 0.68) and HNF1A (r = 0.61).
  • The study utilized a computational pipeline to analyze the F7 promoter regulatory landscape and simulate potential transcriptional enhancements.
