Two in vitro methods are presented to compare the specificity of first- and next-generation CRISPR/Cas9 systems.

  • The methods allow for evaluation of the specificity of different guide RNA/Cas9 complexes on known off-target DNA sequences.
  • One assay provides a broad specificity profile by assessing cleavage across a large library of potential off-target DNA sequences.
  • These assays may facilitate the assessment of CRISPR/Cas9 systems on any DNA target sequence efficiently.
