Genome editing inside living organisms using a new Cj4Cas9 tool

Essence

Cj4Cas9 is a compact CRISPR nuclease that edited mouse embryos and mouse liver, with an engineered version expanding its targeting range.

Evidence

This genome-editing platform study screened 11 CjCas9 orthologs, tested Cj4Cas9 in GFP assays, mouse zygotes, and AAV8-delivered mouse liver editing of Pcsk9, and engineered enCj4Cas9 with L58Y/D900K mutations.

Caveat

The evidence is preclinical and platform-focused, so therapeutic usefulness in humans is not established.
