Interleukin-2 helps improve CD4+ T cell memory by increasing cells that respond to IL-7

Abstract

IL-2 is identified as a key factor influencing memory T cell survival by regulating IL-7 receptor expression.

  • IL-2 receptor alpha is expressed early in T cell activation, while IL-7 receptor alpha expression is lost at this stage.
  • Later in the immune response, IL-7 receptor alpha is reexpressed as IL-2 receptor alpha is silenced.
  • This reciprocal expression pattern is disrupted in CD4(+) T cells lacking IL-2 signals.
  • Primed T cells deficient in IL-2 or IL-2 receptor alpha can activate normally but fail to reexpress IL-7 receptor alpha.
  • The inability to reexpress IL-7 receptor alpha in IL-2 deficient T cells leads to poor development of long-lived memory T cells.
  • Introducing IL-7 receptor alpha into IL-2 deficient T cells can restore their long-term survival capacity.
