Using Immune-Boosting Nanoparticles to Change Fat Tissue and Encourage Fat Burning to Help Treat Obesity

Abstract

IL-4-loaded lipid nanoparticles (IL-4/LNP) achieved over 70% encapsulation efficiency and reduced weight gain by approximately 10% in a high-fat diet-induced obese mouse model.

  • IL-4/LNP demonstrated superior reprogramming of M1 macrophages to M2 compared to free IL-4, indicated by increased CD206 and decreased inflammatory markers.
  • In co-cultures of adipocyte-mimicking cells and macrophages, IL-4/LNP-polarized M2 macrophages led to significant reductions in lipid droplets and promoted beiging of adipocytes.
  • Localized injections of IL-4/LNP in obese mice resulted in an increase in multilocular beige-like adipocytes and upregulation of thermogenic genes like Ucp1 and Pgc1α.
  • IL-4/LNP treatment was associated with reduced levels of inflammatory cytokines such as IL-6 and showed improvements in liver fat accumulation without systemic toxicity.
