Intestinal Immune Cells Package Peptides in Tiny Carriers to Create Long-Lasting Oral Treatments

Updated

Abstract

A single oral dose of EXE-COS@PCC/SA achieved up to 8 days of hypoglycemic effects in db/db mice.

  • The oral delivery system utilizes porous calcium carbonate/sodium alginate microparticles to enhance the absorption of exenatide-chitosan oligosaccharide.
  • Exenatide can be selectively internalized by intestinal macrophages, promoting their shift to anti-inflammatory M2-phenotypes.
  • M2-macrophages facilitate the repackaging of exenatide into exosomes, which are then transported to sites of inflammation in diabetes.
  • Exosomes target the pancreas, liver, and adipose tissue, contributing to the restoration of tissue balance and glycemic control.
  • The system shows potential for long-term management of type-2 diabetes and may improve systemic inflammation and lipid metabolism.
