Muscle injection followed by nasal spray of mRNA vaccine triggers protective mucosal immunity against COVID-19 in mice

Abstract

Intranasal immunization alone induced weak systemic antibody responses and failed to elicit mucosal immunity.

  • An 'intramuscular prime and intranasal boost' regimen provoked strong systemic and mucosal immunity.
  • Significantly elevated levels of antigen-specific T cells and mucosal IgA were observed following the prime-boost strategy.
  • Intranasal boosting promoted IgA class switching in antigen-specific B cells.
  • The B cell receptor repertoire became more diverse with the optimized immunization approach.
  • This approach for mRNA vaccines encapsulated in lipid nanoparticles may enhance mucosal immunity against viral infections.
