Nasal delivery of gene-editing nanoparticles targeting MAPK9 may reduce brain inflammation after injury

Abstract

Intranasal delivery of Iba-1-targeted CRISPR-LNPs resulted in effective MAPK9 modulation in Iba-1+ myeloid cells within the injured brain.

  • Targeting MAPK9 in myeloid cells may reduce pro-inflammatory activation, as indicated by decreased levels of iNOS, NLRP3, CD80, and CCL2.
  • Increased expression of anti-inflammatory markers CD206 and Arg1 was observed following CRISPR-mediated MAPK9 targeting.
  • In a mouse model of TBI, Iba-1-targeted CRISPR-LNPs showed preferential association with Iba-1+ cells over NeuN+ neurons.
  • Treatment led to reduced microglial/macrophage activation and diminished pro-inflammatory cytokine expression in the peri-contusional cortex.
  • A favorable safety profile was noted, with no detectable toxicity in major organs examined.
