Nasal delivery of Netrin-1 mRNA in tiny vesicles may reduce brain cell death after stroke

Updated

Abstract

Intranasal administration of Netrin-1 modRNA-loaded extracellular vesicles reduced infarct volume by 40% in a mouse model of ischemic stroke.

  • Netrin-1 modRNA is delivered effectively to neurons via extracellular vesicles derived from human umbilical cord mesenchymal stem cells.
  • In vitro experiments showed that EV@mNetrin-1 significantly decreased neuronal cell death and improved survival after oxygen-glucose deprivation.
  • In vivo, the treatment led to increased Netrin-1 levels in the ischemic brain tissue and improved neurological outcomes in mice.
  • The neuroprotective effects are associated with the activation of specific signaling pathways in neurons.
