Detecting charged fats in horse blood to monitor mRNA drug use

Abstract

A detection method for ionisable lipids in equine plasma achieved limits of detection as low as 5 pg/mL.

  • Lipid nanoparticle (LNP)-mRNA formulations have shown effective delivery in vivo.
  • A simple method was developed using supported liquid extraction followed by liquid chromatography-high-resolution tandem mass spectrometry.
  • Isopropanol and heptane were used to dilute plasma, improving the recovery of lipids.
  • The method successfully detected ALC-0315 in plasma samples from a horse treated with LNP-mRNA.
  • This represents the first method for screening ionisable lipids in equine biological samples, aiding in doping control.
