Abstract

In an endotoxemia mouse model, TRI LNPs reduce inflammatory cytokines by approximately 4-fold compared to SM-102 LNPs.

  • Lipid nanoparticles (LNPs) are effective for delivering nucleic acids but can provoke unwanted immune responses.
  • Incorporating FDA-approved corticosteroids into LNP formulations creates steroid LNPs with adjustable anti-inflammatory effects.
  • Triamcinolone (TRI) was identified as the leading steroid, maintaining essential characteristics with 50% cholesterol substitution in SM-102 LNPs.
  • Optimal steroid and cholesterol substitution ratios vary depending on the type of ionizable lipid used.
  • In a multiple sclerosis mouse model, TRI LNPs not only reduced inflammatory cytokines by approximately 3-fold but also provided protection against paralysis.
