Comparing How Sodium Citrate Buffers Affect Lipid Nanoparticles Carrying Circular or Linear RNA

Abstract

High-concentration sodium citrate compromised the in vivo expression of circRNA lipid nanoparticles (LNPs) but not mRNA LNPs.

  • CircRNA demonstrated comparable physicochemical characteristics and in vitro transfection efficiency to mRNA when using the same sodium citrate buffer concentration.
  • LNPs containing circRNA showed lower expression efficiency compared to those containing mRNA when formulated with high-concentration sodium citrate.
  • MC3-LNPs exhibited lower RNA expression efficiency in mice compared to SM-102 LNPs, particularly for circRNA.
  • High-concentration sodium citrate buffer maintained the integrity of both mRNA and circRNA after multiday storage at room temperature.
  • The findings suggest that different LNP formulations may require customized optimization for effective delivery of mRNA versus circRNA.
