Abstract

IscB proteins may use a single noncoding RNA for RNA-guided cleavage of double-stranded DNA.

  • IscB is a putative nuclease related to the ancestors of the RNA-guided endonuclease Cas9.
  • Evolutionary analysis and RNA sequencing were utilized to understand the evolution of CRISPR-Cas9 systems from IS200/IS605 transposons.
  • IscB can be harnessed for genome editing in human cells.
  • TnpB, another protein from the IS200/IS605 family, exhibits RNA-guided nuclease activity and is likely an ancestor of Cas12 endonucleases.
  • A new class of transposon-encoded RNA-guided nucleases, named OMEGA, is proposed.
