Ketamine reduces loss of pleasure across species shown by a reward-based test

Abstract

Ketamine significantly increased response bias toward the more frequently rewarded stimulus in individuals with treatment-resistant depression and rats exposed to chronic stress.

  • A single dose of ketamine (0.5 mg/kg) improved reward responsiveness in individuals with treatment-resistant depression.
  • Rats receiving ketamine (10 mg/kg) also showed increased response bias toward more frequently rewarded stimuli after chronic stress.
  • Both species reached levels of reward responsiveness comparable to healthy controls 24 hours post-ketamine administration.
  • Exploratory analyses indicated that the effect was strongest in humans with higher levels of baseline anhedonia.
  • Ketamine did not affect overall task performance, suggesting it specifically enhanced reward learning.
