Creating a highly sensitive CRISPR-based sensor that avoids signal leaks by using a split activator triggered by self-cleavage for detecting tiny amounts of microRNA

Updated

Abstract

The developed biosensor achieves sensitive miRNA detection with a low detection limit and excellent specificity.

  • MicroRNAs are important biomarkers for cancer diagnosis and prognosis but are difficult to detect due to their low abundance and short sequences.
  • A novel biosensing platform combines catalytic hairpin assembly with dual CRISPR/Cas12a amplification for improved detection of miRNAs.
  • This platform decouples target recognition from signal output, which reduces nonspecific background signals.
  • The cascade design enables sequential signal amplification, resulting in enhanced signal-to-noise ratios.
  • The biosensor demonstrated satisfactory accuracy and recovery in practical applications, including cell lysates and human plasma samples.
