Functional inhibition of let-7a-5p mitigated S1-induced inflammatory cytokine release and markers of pyroptosis.

  • Dysregulated microRNA expression, including let-7a-5p, was identified in response to SARS-CoV-2 spike S1 stimulation.
  • The let-7a-5p microRNA is associated with regulating neuroinflammation in human microglial cells.
  • SHIP-1 was established as a direct target of let-7a-5p through validation using a luciferase reporter assay.
  • Upregulation of TLR3, TLR2, and TLR4 genes occurred in S1-stimulated microglia, indicating potential involvement in neuroinflammatory processes.
  • Findings suggest a role for the let-7a-5p/SHIP-1 axis in regulating inflammatory responses in microglial cells.
