Testing many lipid nanoparticles to find the best ones for delivering CRISPR gene-editing tools

Updated

Abstract

An optimal lipid nanoparticle candidate achieved approximately 90% EGFP disruption in cells at a concentration of 30 nM.

  • The developed library of ionizable lipids led to the identification of LNPC12A9 as an effective delivery platform for CRISPR-Cas9 RNP.
  • LNPC12A9 demonstrated consistent editing performance at a low RNP concentration of 15 nM across multiple hepatic cell lines.
  • Coencapsulation of RNP and a single-stranded oligodeoxynucleotide template resulted in a homology-directed repair rate of 37.8%.
  • Systemic administration of LNPC12A9 in mice resulted in a liver-targeted disruption rate of 1.39% for the Pcsk9 gene.
