More mRNA inside lipid nanoparticles increases gene delivery effectiveness

Updated

Abstract

The two-step dialysis process increases mRNA loading per lipid nanoparticle (LNP) and improves transfection potency.

  • A two-step dialysis method was tested, starting with formulation at pH 4 and followed by fusion induction at the same pH.
  • This protocol maintained ethanol concentration at 25% during the fusion step.
  • The new method led to a decreased proportion of empty LNPs compared to the traditional one-step process.
  • In vitro and in vivo transfection potencies were improved with the two-step protocol.
