Lipid nanoparticles with zwitterionic lipids improve mRNA delivery efficiency

Abstract

Essence

Replacing DSPC with zwitterionic zTOT lipids made lipid nanoparticles more efficient mRNA delivery platforms in these experiments.

Evidence

Formulation and platform experiments comparing zTOT LNPs with DSPC-LNPs found TOT-15 gave 5-fold higher gene expression, almost 100% knockout without toxicity, and calculated endosomal escape above 9.6%.

Caveat

The work remains a delivery-platform study, so it does not establish therapeutic efficacy or clinical safety.
