Liraglutide improved insulin sensitivity and decreased fasting and postprandial glucose levels after 2 weeks, independent of weight loss.

  • Liraglutide led to significant weight loss, while the dipeptidyl peptidase 4 (DPP-4) inhibitor sitagliptin did not cause weight changes.
  • Insulin sensitivity improved with liraglutide as measured by multiple indices, even before significant weight loss occurred.
  • Sitagliptin increased levels of endogenous GLP-1 and GIP but did not enhance insulin sensitivity or reduce fasting glucose levels.
  • Diet-induced weight loss improved insulin sensitivity through specific measures but did not lead to reductions in glucose levels.
  • The GLP-1 receptor antagonist exendin(9-39) reversed the insulin sensitivity improvements seen with liraglutide.
