Liraglutide helps obesity-related type 2 diabetes by changing immune cell cholesterol use and gene activity to boost anti-inflammatory cells and fat burning

Updated

Abstract

Liraglutide treatment in O&T2DM mice resulted in significant reductions in body weight, fasting blood glucose, and HOMA-IR.

  • Liraglutide decreased visceral fat accumulation and promoted adipose tissue browning in a mouse model of obesity-related type 2 diabetes.
  • The treatment increased the proportion of M2 macrophages in adipose tissue and activated the cholesterol efflux pathway associated with LXRα/ABCA1.
  • In vitro, liraglutide suppressed M1 macrophage polarization and upregulated M2-type markers, enhancing cholesterol metabolism-related gene expression.
  • Histone modification analysis revealed that liraglutide increased levels of H3K27ac in the promoter regions of genes related to M2 polarization and cholesterol metabolism.
  • Co-culture experiments indicated that liraglutide-reprogrammed macrophages promoted adipocyte thermogenic gene expression and mitochondrial activity through paracrine signaling.
