Sensitive detection of multiple liver cancer microRNAs using a logic-controlled CRISPR test with boosted signals

Abstract

Detection limits of 78.88 fM for miRNA 122 and 65.26 fM for miRNA 223 were achieved using a novel biosensing platform.

  • The platform integrates a logic gate circuit with an engineered CRISPR-Cas system for early hepatocellular carcinoma screening.
  • Multiple miRNA inputs must co-recognize to generate a unified DNA output, which activates the CRISPR-Cas12a system.
  • Enhanced multi-legged crRNA and a cube-based framework probe improve the system's reaction kinetics and activity.
  • Clinical validation demonstrated excellent correlation with RT-qPCR (all R > 0.98) using 36 samples.
  • Areas under the ROC curves of 0.8514 and 0.9244 effectively distinguish liver cancer patients from healthy individuals.
