A high probability of loss-of-function intolerance (pLI = 0.96) is observed for the human brain-specific TAU isoform 0N3R.

  • The 0N3R isoform of TAU is essential for proper neurodevelopment in humans.
  • Loss of this isoform is linked to neurodevelopmental impairments, potentially leading to intellectual disability.
  • In contrast to mice, where other proteins can compensate, loss of TAU in human neurons disrupts neurite outgrowth and axon formation.
  • Functional redundancy among the six brain-specific TAU isoforms allows rescue of developmental deficits when any isoform is re-expressed.
  • The scarcity of loss-of-function mutations in 0N3R suggests strong evolutionary pressure to maintain its function.
