Mechanical stimulation of bone marrow stem cell vesicles promotes bone-forming cell growth and development by activating the Wnt/β-catenin pathway

Abstract

Exosomes derived from mechanically stimulated bone marrow stem cells (MS-Exo) more effectively reversed the effects of glucocorticoids on bone cell behavior compared to conventional exosomes.

  • MS-Exo significantly improved cell proliferation, osteogenic differentiation, and mineralization in osteoblasts affected by glucocorticoids.
  • Transcriptomic analysis indicated significant activation of the Wnt/β-catenin signaling pathway in cells treated with MS-Exo.
  • Validation experiments showed increased levels of β-catenin, LRP6, and TCF7 following MS-Exo treatment.
  • The Wnt pathway inhibitor Dkk-1 partially reduced the pro-osteogenic effects of MS-Exo, suggesting a role for this pathway in its mechanism.
