Using microfluidics to create solid fat nanoparticles that improve delivery of semaglutide

Updated

Abstract

The formulation with a semaglutide to DOTAP molar ratio of 1:18 demonstrated particle sizes of less than 300 nm and nearly complete encapsulation efficiency.

  • Semaglutide is a peptide widely used for managing type 2 diabetes and obesity but faces challenges in oral delivery due to gastrointestinal instability and poor permeability.
  • The combination of hydrophobic ion pairing with DOTAP and solid lipid nanoparticles was explored to enhance the delivery properties of semaglutide.
  • Formulation F10, with a 10% peptide concentration, showed good stability under simulated gastrointestinal conditions and a sustained-release characteristic.
  • This formulation interacted strongly with mucus while still being able to diffuse through the mucin network.
  • Cytocompatibility studies indicated acceptable cell viability at relevant concentrations.
  • Permeability experiments indicated an approximately 6-fold increase in apparent permeability compared to free semaglutide.
