Abstract

APOE4 fibroblasts exhibited impaired mitophagy and significantly reduced lysosomal proteolytic activity.

  • Mitochondria in APOE4 fibroblasts showed increased markers for mitochondrial content but reduced respiratory capacity and elevated proton leak.
  • Excessive mitochondrial reactive oxygen species (ROS) were observed in APOE4 cells, indicating oxidative stress.
  • Defective autophagic flux was noted in APOE4 fibroblasts, as evidenced by reduced colocalization of autophagy-related proteins.
  • Lysosomal activity was significantly lower in APOE4 cells and did not improve under starvation conditions, unlike sAD cells.
  • Pharmacological intervention targeting mitochondrial ROS restored lysosomal degradation and mitochondrial respiration in APOE4 fibroblasts.
