Abstract

Overexpression of MFB1 can suppress cell growth defects caused by mutations in mitochondrial protein import.

  • Mitochondrial biogenesis requires importing 1,000-1,500 proteins through TOM and TIM complexes.
  • Mutations in ANT1 can lead to diseases by clogging these protein import pathways and causing mitochondrial Precursor Overaccumulation Stress (mPOS).
  • Overexpressing MFB1 helps counteract growth defects from a clogger allele of AAC2, the yeast version of ANT1.
  • Disrupting MFB1 worsens cell growth issues under clogging conditions and increases mitochondrial proteins stuck in the cytosol.
  • Mfb1 may interact with proteins like Tom22 that are part of the TOM complex, impacting mitochondrial import.
  • Loss of MFB1 activates genes for cytosolic chaperones, including HSP31, which could protect against mPOS.
